Limited DNase I digestion of isolated nuclei was used to probe and map accessible regions of the fly genome. The resulting purified fragments were sequenced using an Illumina NGS Platform, and the short sequence reads were aligned to the reference genome. Clusters of aligned sequence tags were used to define both highly accessible regions of the genome as well as DNase I hypersensitive sites (DHSs).
